Ok I don't have IS250 but I have Mark X 250G which is basically the same car technically but just different chassis. So to try answer your question:
1) It'll be just a bit more expensive but not quite like BMW/Mercs level yet. Can also already find parts in chop shops. It's still a Toyota after all so not too different than maintaining say a Camry 2.5.
2) Not that I'm aware of. It's technically a pretty reliable car without any nagging issues.
3) Yes, the same 4GR-FSE engine, same gearbox, same brakes, and shares many suspension parts too. However, compared to Mark X first gen (GRX120), IS250 power is slightly down tuned (215ps in GRX120 vs 205ps in IS250), but if compared to subsequent gen Mark X then already the same 205ps.
4) I'd actually suggest you to buy Mark X GRX120 instead coz it's cheaper but yet more spacious/bigger. But between IS205 vs Camry 2.5, surely IS250 because at least it's a bit faster and looks way better still.

Hope this helps even though my response is more based on my ownership of Mark X for almost 2 yrs now.
